Line is to use the internal preamp when receiver has no phono input. Phono is for use with a receiver that has a phono input.

The preamplifier raises the signal level to line level, and it applies the RIAA equalization curve that is required for the playback of vinyl records. This allows you to use the turntable with devices that do not have a dedicated PHONO input for a turntable on them. Connect the turntable to any line level input, such as the AUX or CD input. If the device DOES have a PHONO input on it, there is a phono preamplifier built into the device. Only one phono preamplifier should be used at a time.
