Yes. It will. On the rare occasion that my battery runs low, I plug it into the USB charger in my car and continue listening to my podcasts. Please note that if it is plugged into your car's charging system, it will automatically turn on when you start your car. There may be a way to turn this off in the settings, but it hasn't been a problem for me, so I haven't looked.

However, if in car and hooked to power for charging AND at same time using the AUDIO IN wire to your car stereo, you may hear some static noise while playing songs. This is the nature of hooking usb power to ANY device used to play audio through an audiojack wire into the car stereo. My smartphone even does it. But yes it is possible to play at same time as charging.
